Enabling NGS Multiplexing
FactorLS™ 24 and 96 Unique Dual-Indexed (UDI) Stubby Adapter Kits are intended for use with NGS libraries created using End Repair / A-Tailing followed by TA ligation. Ligated products are then PCR amplified using the supplied Indexing Primers to create an Illumina TruSeq compatible library that can be sequenced on Illumina sequencing platforms.
Duplexed Stubby Adapters are provided in a 2 mL screw cap tube with volumes supporting 24 or 96 reactions. The Indexing Primers are provided in 96 well plates (available in 24 and 96 reactions) and sealed with pierceable foil seals. Each well contains a Unique Indexing Primer pair intended for a single use and contains both the forward (p5) and reverse (p7) primers with UDI at a concentration of 10 µM each. Each plate is organized by column, with the 24 reaction plates containing Indexing Primers in Columns 1-3.
Forward compatible Illumina instruments include: NovaSeq® 6000 (v1.0 reagent kits); HiSeq® 2000, 2500; NextSeq® 1000, 2000 (using v2 Sample Sheet); MiSeq®; MiniSeq® (rapid reagent kits)
Reverse compatible Illumina instruments include: NovaSeq® 6000 (v1.5 reagent kits); HiSeq® 3000, 4000, X; NextSeq® 1000, 2000 (using v1 Sample Sheet); NextSeq® 500, 550; MiniSeq® (standard reagents); iSeq® 100
