Overview
Sequencing primers are short oligonucleotides that initiate DNA synthesis during sequencing reactions for the application of next-generation sequencing (NGS).
Prior to sequencing, genomic libraries are prepared by attaching adapter sequences to the ends of DNA fragments to allow for binding of the libraries to the sequencing platform, binding sites for sequencing primers and even multiplexing for sample identification. NGS primer and adapter sequences must be carefully designed and optimized to reduce off-target annealing and amplification, and any added chemical modifications must be adequately removed prior to use in downstream applications.
