Antisense Oligonucleotides (ASOs) & Gapmers for Targeted Therapy Development

ASOs are short, synthetic single-stranded DNA or RNA molecules designed to selectively bind to messenger RNA (mRNA) and modulate gene expression.

This binding can either block the translation of the mRNA into protein or trigger its degradation, effectively “silencing” the target gene. This mechanism of action makes ASOs invaluable in treating a wide range of diseases, from rare genetic disorders to cancers, and must be manufactured with high purity and accuracy to ensure specificity. ASOs may also require the addition of complex modifications to enhance binding and stability for improved efficacy.

Gapmers are specialized ASOs designed with a central DNA “gap” flanked by chemically modified nucleotides. This unique structure enhances target binding affinity, nuclease resistance, and RNase H recruitment, making gapmers powerful tools for gene silencing and modulation.
