Primers are short oligonucleotides that serve as starting points for DNA synthesis, enabling the amplification of genetic material in techniques like polymerase chain reaction (PCR).

In addition to amplification, quantitative PCR (qPCR) utilizes DNA probes labeled with fluorescent dyes to measure the amount of DNA present in real-time. Primer and probe sequences must be carefully designed and optimized to reduce off-target annealing and amplification, and any added chemical modifications must be adequately removed prior to use in downstream applications.
